Default Diagnostic port problem!!! HELP

I have a 2000 C5 Corvette that a friend of mine and I have been tuning. Until today everything was going well. I wanted to drive my car and data log a little so I took off for a drive. I found that when I plug my HP Tuners in, it says that the interface is found but the vehicle has not, so I reconnected and got the same message. So I decided to plug in my handheld programmer (SuperChips) which as of last night was working just fine, and that couldn't get a connection either. Any advice on what to look for? I don't want to buy another PCM, I think the problem is in the port itself. Is that replacable or fixable?
